Did you know that the daffodil (or Påskliljan in Swedish) is the flower most associated with Easter in Sweden?
After a long winter, the streets have already been filled with little flowers of different colours all over the grass and they bring a lot of joy (apart from allergies ;)).

Highly accurate diluent volume

We control our dilution cups very much to make sure they have as little deviation from 9 mL as possible. We regularly make statistics of our analyses, and this is the latest one we have done.
🔊EXPLANATION:
In the following graph you can see the amount of Dilucups classified according to 3 different volume intervals during the last 3 years:
9 mL - (1 to 0,4999 %) ➡ 8,91 mL to 8,955 mL
9 mL ± 0,5 % ➡ 8,955 mL to 9,045 mL
9 mL + (0,5001 to 1 %) ➡ 9,045 mL to 9,09 mL
❕Note: according to ISO standards the volume must be: 9 mL ± 2 % (8,82 mL to 9,18 mL)
Results are based on approximately 15.000 volume measurements per year.
📈 RESULTS:
In 2021, 99,97 % of Dilucups had a volume of 9 mL ± 1 %, of which 89,21 % were in the range of 9 mL ± 0,5 %.
In 2022, 99,9% had a volume of 9 mL ± 1 %, of which 84,45 % were in the range of 9 mL ± 0,5 %.
Last year, 99,92% had a volume of 9 mL ± 1 %, of which 91,72 %* were in the range of 9 mL ± 0,5 %.
*This increase compared to 2022 is not a coincidence. Since summer 2023 we have been working harder so that the volume is in the range of 9 mL ± 0,5 %. This is because we know how important it is to have a highly accurate volume for many industries, such as probiotics.
📝 CONCLUSION:
Almost all Dilucups have a volume of 9 mL ± 1 %, with normal values being in the range of ± 0,5 %.
The results are consistent over the years, and our goal is to continue increasing the percentage of Dilucups that have a volume of 9 mL ± 0,5%.

You can safely use micropipettes with Dilucup®

Serial dilution with tubes and a micropipette can be challenging because the risk of contaminating the inside of the tube with the pipette is high. That is why straws are often used, but how can you be sure about the accuracy of the pipetted volume?
The good thing about Dilucup is that since it is a low cup, you can safely use a micropipette and get the best possible volume accuracy.

The most important thing when performing serial dilutions is that the volume is accurate. Otherwise, you can forget about having a good result because the accumulated error will give you a wrong enumeration of microorganisms.
Volume statistics (based on 25.000 measurements):
- 94,4 % -> 9 mL ± 0.5 %
- 99,95 % -> 9 mL ± 1 %
- 100 % -> 9 mL ± 2 % (according to ISO standards)
Methods where filling tubes or other recipients is done in the laboratory cannot guarantee a volume as accurate as Dilucup.

Latest volume accuracy analysis

The volume of a diluent must be accurate to have reliable results and according to EN ISO 6887-1: 2017 the volume should not deviate from 9,0 mL ± 2 % (9 mL ± 0,18 mL).
We want to be better than this and that is why Dilucup® aims for a volume range of 9 mL ± 1 %.
99,95% of the 25.000 latest Dilucups analysed in our QC laboratory had a volume of 9 mL ± 1 %, with an average of 9 mL + 0,26 % mL (9,02 mL) and a SD of 0,21 %.

How to enumerate microorganisms?

How to enumerate the microorganisms in a food sample using Dilucup?
- First homogenize the sample if it is solid. This is the first 1/10 dilution.
- Once the sample is suspended or if the sample is liquid, serial dilute it using Dilucup.
- Plate 1 mL of the selected suspension(s) and incubate.
- Count the colonies (the plates should have between 30 and 300 CFUs to get a statistically valid result).
- Divide the average number of CFUs per the dilution level to get the number of microorganisms per mL or g in the original sample.
